Constitutional Chips 4.0 (PerkinElmer®)
DescriptionComparative Genomic Hybridization (microarray-CGH) is a technology based on detection of genomic imbalance and chromosomal aberrations (abnormalities of a few hundred kilobases up to several megabases). This technology allows quantitative analysis of genome abnormalities with a a resolution of 50 to 100 times higher than with constitutional karyotyping. Hybridization & Labelling Reagents Kits content: ![]() • 1 vial (495 µl) of Hybridization Buffer 1:
Hybridization Buffer 1 contains human Cot-1 DNA,sheared salmon testis DNA, and NaCl.
• 1 vial (330 µl) of Hybridization Buffer 2:
Hybridization Buffer 2 contains SSC (Saline Sodium Citrate), formamide and Dextran.
Warning: Formamide is toxic. Use with caution. • 1 vial (1200 µL) of aCGH Water: aCGH Water is RNAase/DNAase-free water. 2. Labelling reagents: • 1 vial (140 µl) of labelling buffer:
This buffer contains dATP, dCTP, dGTP, dTTP on base of Tris-HCl buffer. • 1 vial (425 µl) of Random primers: This buffer contains random primer oligonucletides on base of Tris-HCl buffer. • 1 vial (55 µl) of Klenow: This buffer is a DNA polymerase. • 1 vial (45 µl) of Cyanine 3: This vial contains dCTP-Cy3 on base of Tris-HCl buffer. Warning: keep the vial protected from light. • 1 vial (45 µl) of Cyanine 5: This vial contains dCTP-Cy5 on base of Tris-HCl buffer. Warning: keep the vial protected from light. All Hybridization and Labelling reagents must be stored at -30/-16°c. Expiry date is stated on the vial label. 3.Wash Buffer Pack: • Prewash Buffer : SSC 2X, SDS 0,1% 40 ml for 10 slides
• Wash Buffer 1: SSC 1X, SDS 0,1% 400 ml for 10 slides • Wash Buffer 2: SSC 0,1X, SDS 0,1% 200 ml for 10 slides • Wash Buffer 3: SSC 0,1X 400 ml for 10 slides • Rinse Solution: Isopropanol 200 ml for 10 slides All solutions are stored at room temperature. |
